Yes. fellow your attched configure image, kit would enter psoc 4100plus mode, and the creator4.2 could connect the kit and program. Howerver. change the switch to EZ-BLE mode. then the error prompt would happend when creator4.2 was trying to connect this kit. thanks.

You need to create two projects in PSoC Creator, one for programming PSoC 4100S plus module and other for programming PRoC BLE Module. For programming PSoC 4100S Plus you need to switch the module to PSoC 4100S mode and for programming PRoC BLE Module switch to EZ-BLE Mode.
For creating the project for PRoC BLE module, In the PSoC Creator select Target Module and select CYBLE-022001-00.
